HealthylifeOrlistat is used to treat obesity and help in weight loss. Orlistat is to be used along with a low-calorie diet and regular physical exercise. Obesity is a common condition associated with excessive body fat, which in turn increases the risk of other health problems such as diabetes, high blood pressure, certain cancers, and heart disease. Intake of more calories than you burn by exercise or daily activities results in obesity.
Orlistat contains Orlistat, which works in the small intestine and stomach and prevents the action of enzymes that break down fat, which is absorbed by the body. Orlistat decreases the absorption of fat from the food you eat. Thus, fat passes through the gut and is excreted in the faeces, which makes the body unable to use fat as a source of energy or convert it into fat tissue. Thereby, Orlistat helps in weight loss.

Objective:To compare the efficacy of orlistat versus orlistat plus oral methotrexate in the treatment of overweight and obese patients with type 2 diabetes. Methods: This was a randomized, double-blind, placebo-controlled, parallel-group, double-dose, parallel-group study of patients randomized to orlistat or to receive methotrexate for the treatment of overweight and obese patients with type 2 diabetes.
Setting:Adult patients with type 2 diabetes (diabetic patients with overweight and obesity) who had not previously received oral methotrexate and were eligible for the study if they: (i) were treated with orlistat, or (ii) had a body mass index of greater than 27 kg/m2 for the first 12 weeks, or a body mass index of greater than 30 kg/m2 for the last 12 weeks, or a body mass index greater than 28 kg/m2 for the last 12 weeks, or a body mass index greater than 30 kg/m2 for the first 3 months.
Participants:6% of the patients in the orlistat group and 1% in the orlistat plus methotrexate group (n=6).
Intervention:The patients were randomized to receive orlistat or methotrexate (2.5 mg/day) for the treatment of overweight and obese patients with type 2 diabetes. After 12 weeks, the patients were randomized to receive methotrexate (5 mg/day) or oral orlistat plus methotrexate (1 mg/day) for the treatment of overweight and obese patients with type 2 diabetes.
Main outcome measures:Baseline clinical assessments and adverse events.
Results:The proportion of patients with clinical data at 12 weeks was <0.001 for patients treated with orlistat and <0.001 for patients treated with orlistat plus methotrexate. Patients treated with orlistat had a lower incidence of adverse events, including gastrointestinal symptoms and weight gain. Patients treated with orlistat plus methotrexate were more likely to have more serious side effects, including gastrointestinal symptoms and weight gain, than patients treated with orlistat alone (23% vs. 7%, respectively; p=0.004). There was no difference in the incidence of other serious adverse events between the 2 groups.
Conclusion:The efficacy of orlistat was not statistically significantly different from orlistat plus methotrexate (3-year placebo-controlled study).
